Hit and Lead Finding Libraries
The focus for our library and building block design is to provide compounds and reagents suitable for generating Hit Finding and Fragment Based Discovery (FBD) libraries. Our design work is focussed on saturated ring systems (or saturated rings linked to heterocycles) since we believe that saturation confers increased novelty, aqueous solubility, three dimensional complexity, and a favorable balance between hydrophilicity and lipophilicity.
Quality Control: We provide CoAs [and MSDS] with each compound and retain analytical samples which we periodically re-analyze to confirm shelf-life. CoAs confirm purity exceeding 95% (area) by HPLC. 1H NMR is used to quantify residual solvent or water content for each compound. Structural confirmation is achieved by LC-MS and 1H NMR.
Physical Chemistry: We routinely determine equilibrium solubilities (HPLC) and empirical logP values for two compounds per library. The compounds are selected based on the expectation of their having the highest and lowest aqueous solubility and logP, per individual library, in order to obtain a range for each library.
